According to the National Sleep Foundation, a sleep disorder which affects approximately 1 in 2,000 people. In 1986, the Food and Drug Administration (FDA), approved for Modafinil to be used as an experimental treatment for narcolepsy. Since then, its effective uses have expanded over the years to treat several other sleep and fatigue-related disorders.

Today, based on its proven cognitive-enhancing and motivation-boosting benefits, Modafinil has become one of the most popular Nootropics. The military, students, business people, and athletes are some of the many people all over the world who take Modafinil. And, that is to name only a few. How Does Modafinil Work?

Modafinil works by selectively activating neurons and primarily focuses on stimulating the brain’s amygdala and the hypothalamus.

The effects of using Modafinil are more or less similar to those associated with other conventional stimulants. Take for example the amphetamine Adderall. Similar to Modafinil, Adderall is a psychostimulant designed to produce short bursts of psychomotor activity (i.e. physical movement, coordination, and dexterity) to help treat narcolepsy and ADHD. While its actual mechanism of action has not yet been fully explored, it is, however, clear that the effect Modafinil has on several significant neurotransmitters is at the core of its potency. Research studies, for example, show Modafinil can modulate the brain’s orexin systems. Meaning it can change the levels of dopamine, serotonin, norepinephrine, and histamine in your body. The following is a brief outline of how Modafinil effects certain functions of the body.

Glutamate

Modafinil elevates the level of glutamate which is the principal excitatory neurotransmitter in the brain. It does this by stimulating the extracellular expulsion of glutamate within the brain’s hypothalamus. This simply means as glutamate is largely responsible for sending signals between nerve cells, it plays an important role in brain function such as learning and memory. Norepinephrine

Some research suggests it could be directly related to the action on the dopamine receptors and orexin system. While other research, conducted by Paul Gerrard and Robert Malcolm, suggests that it may be due to the way in which “Modafinil increases cortical GABA through a norepinephrine-mediated pathway.” If accurate, this would mean modafinil helps to increase gamma-aminobutyric acid (GABA) uptake but does not directly affect it.

Dopamine

Dopamine is an important neurotransmitter that is directly linked to the stimuli/reward response. It’s a chemical released by neurons in the brain which helps to increase focus and motivation and improve mood. Other stimulants and amphetamines directly stimulate the release of dopamine increasing their levels in an amplified and very rapid manner. Modafinil, however, gradually works through what is referred to as ‘blockage’ of the receptors that expel dopamine from the associated synapses. Thus, effectively elevating the levels of dopamine made available for use in the brain somewhat immediately. This indirect and gradual action on the dopamine system is a significant feature that shows how Modafinil works since it reduces the kind or euphoria associated with addiction.

Histamine

Most of us have heard about histamine in relation to local immune responses and allergies (i.e. hay fever). But, did you know, histamines also perform a significant role with regards to the regulation of the sleep-and-wake cycle? That’s right. The action of Modafinil on the brain’s hypothalamus is what is thought to elevate the levels of histamine. Researchers Brown, Stevens, and Haas support this claim in their research; The Physiology of Brain Histamine. They point out that our histamine levels are “highest during waking and lowest during rapid-eye-movement sleep.” This simply means that a high-level of histamine correlates with a high-level of alertness and vice-versa.

The firing of histamine-releasing neurons, therefore, greatly contributes to our levels of alertness and wakefulness. The effects from the use of Modafinil, however, do tend to wear off. Particularly when the natural neuronal histamine is depleted by an inhibitor that is quite powerful. It runs counter to methylphenidate which reacts in a manner different from the histidine decarboxylase inhibitor—alpha-Fluoromethyl Histidine. If that sounds confusing, just note that too much activation of the histaminergic neurons can cause apoptosis—or cell death. Meaning, while it may increase your intelligence, overuse could be harmful to the health of your brain and body.

Orexin/Hypocretin

Much of the wakefulness-promoting action of Modafinil is attributed to how it acts on the hypocretin, rather orexin system of the brain. Hypocretins are simply neurotransmitters which normally “promote stable wakefulness” according to the Harvard Medical School. Of the billions of neurons in our brain, a relatively small cluster (100,000-200,000) produce hypocretins. These important neurons are situated within the hypothalamus and are responsible for regulating appetite, arousal, and wakefulness. The most common forms of narcolepsy usually result from a lack of hypocretin in the brain. Modafinil addresses this by acting directly stimulating the orexin neurons. As a result, this increases the release of histamines and other neurotransmitters related to alertness and wakefulness.

According to the Harvard Medical School, this is likely due Modafinil's ability to increase the brain’s level of dopamine. Under normal dosage use (100mg - 200mg per 24 hours), Modafinil can help to improve alertness for approximately eight hours. For example, in a study published by Cochrane, researchers Liira et al found Modafinil to be effective at decreasing sleepiness and increasing alertness among shift workers who experience sleep-wake disturbances.

The research also suggests that Modafinil can help to limit us from being prone to distractions enabling us to focus more on the task at hand.
One of the interesting debates which has emerged from the use of Modafinil is its effects on impulse control. Does it help with resisting temptation and keeping us from acting on our urges? Some say—Yes. Researchers from the University of Antwerp and University of Minnesota, for instance, argue in favor of the use of Modafinil to help reduce impulsive behavior of Alcohol-Dependent Patients (ADS) and impulsive gamblers. Other research supports this argument stating that the effects of Modafinil are able to achieve these results by improving the response inhibition and find it to be an effective measure for impulse control. Recent studies which support these claims show that people with Impulse Control Disorder (ICD) have been able to successfully recover through the use of Modafinil. Dr. John Krystal, Editor of Biological Psychiatry suggests that "if you are impulsive, Modafinil may help your self-control."

Provides Neuroprotective Properties

Modafinil can help to promote brain health.
Findings from a 2008 study published by Brain Research, which explores the neuroprotective effects of Modafinil, indicate that Modafinil can actually help to promote brain health. Researchers Van Vliet et al suggests this is largely attributed to the fact that it has antioxidant properties. Chemical agents that help to protect our body from damage caused by free radicals. Modafinil, however, is not an antioxidant. But, it has proven to be capable of reducing the availability of damaging free radicals within the brain tissue. Research conducted by Gerrard and Malcolm, published in the Journal of Neuropsychiatric Disease and Treatment, show that Modafinil’s mechanisms of action have proven antioxidative and neuroprotective effects.

Better Bioavailability

Probably one of the underrated key benefits of Modafinil is its bioavailability. Unlike other smart drugs, Modafinil is proven to be more superior in its ability to be more bioavailable. This means the degree and rate at which Modafinil enters the body’s systematic circulation is greater than many other nootropics.

Modafinil’s bioavailability allows it to better target particular areas and be quickly absorbed through the body’s circulatory system. Due to its fast-acting absorption, according to the EMC, Modafinil’s peak plasma concentrations tend to occur between 2-4 hours. Also worth noting, while taking Modafinil with food has no effect on its bioavailability, it may delay reaching peak concentration by about an hour depending on the solubility of the food taken with it.

Fewer Side Effects

Of the many advantages, Modafinil provides, perhaps the most important is the benefit of having fewer side effects. Especially by comparison to other smart drugs. Besides, what use is a smart drug if it does more harm than good? From the various types of smart drugs I’ve tried, Modafinil has by far had the least side effects, most instances none. The time I did experience the side effects of a headache and anxiousness was when I exceeded the recommended daily dosage.

Although, I think stress and environment also played a part during that time. Naturally, however, some people will experience side effects while others will have none. In fact, studies have found that “Modafinil boosts higher-order cognitive function without causing serious side effects.” Further to having fewer side effects, Modafinil is also known to be better tolerated by the body. This means it has a much lower potential for addiction and abuse by comparison to alternatives such as Adderall.

Where to Buy Modafinil

When I published an earlier version of this post, this section included reviews of several online providers where you can buy Modafinil. I’ve since removed those providers and updated this section due to the following reasons.
  1. Most of the previously reviewed online pharmacies are no longer in business.
  2. I’ve had, let’s say, “less than favorable” experiences with the other providers that turned out to be disappointing and expensive. Therefore, removing them to help you avoid these providers.
  3. The most common question I get asked is: “which provider do you buy Modafinil from?” So, I’ve narrowed it down to who I think is the best Modafinil provider.
A reliable supplier of Modafinil is Afinil Express. In my experience, they offer quality Modafinil at very reasonable prices and offer it from two very reputable pharmaceuticals. They are Modalert (from Sun Pharma) and Modvigil (from HAB Pharma). Sun Pharmaceutical Industries and HAB Pharmaceuticals & Research are both world-leading pharmaceuticals. This is because they both adhere to strict quality control systems and procedures. Meaning you can be assured you are not paying for a poor quality or fake product.

Modalert vs. Modvigil

What is the difference between the two? In my opinion, not much. Both Modalert and Modvigil come in 200mg dosages containing the same active ingredients. While the effects are the same Modalert seems to be the more popular. As the effects vary by individual, you may have to try both to see which works best for you. The only slight difference is in pricing. Generally, per 50 tablets, Modalert tends to be slightly more (c.$1.70 USD) per tablet than Modvigil (c.$1.30 USD). Personally, I use Modalert as I’ve found its effects to be very consistent. It kicks in fast and lasts almost a solid 7 hours. Whether you choose Modalert or Modvigil, I’d suggest with keeping with either one or the other and not mixing the two.

What to Look Out for When Buying Modafinil

Buying Modafinil (off-label) that doesn’t work, is the last thing you want after spending your time and money. Following the tremendous popularity of Modafinil, an influx of new providers have emerged. Some good, some not so good. This has meant, there are providers out there which sell poor quality and fake supplements. Meaning it can be difficult to find a quality provider. So, to help you avoid making costly mistakes, here are some things to look out for to help ensure you get the best quality Modafinil first time and every time. And, to help ensure you are getting a good quality product at a fair price.

Fakes

Producing poor quality and fake Modafinil tablets to sell at a premium or at a cheaper price is a tactic some manufacturers use to make easy money. Unfortunately, fakes can be as accessible and look just as real as any good product. This makes it difficult for customers to tell what is real and what is fake. As a result, cases have been reported whereby manufacturers have been known to fake the medication. For legal reasons, I cannot name any names, however, here are some tips for avoiding fakes.
  • Counterfeit medicine often means one of two things; (1) it’s made of the wrong or no active ingredients or, (2) the right active ingredients have been used but, with the wrong doses. In either scenario, this often makes it very difficult to spot whether or not the Modafinil you are buying is real or fake.
  • Fakes can also be connected to what’s known as a “bad batch.” One way of identifying whether you have bought from a bad batch is simply by, the size of the tablets. If you notice that the tablets are relatively smaller than they should be, it’s likely it’s made from a bad batch and, therefore, regarded as fake.

Cheap Deals

Good quality Modafinil, for 50 tablets, will normally work out to be $1.50-$1.80 (USD) per 200mg.
Cheap or heavily discounted units of Modafinil should always be something to be wary about. If the Modafinil tablets you are looking to buy are rather cheap, check the manufacturer behind who’s producing it. If possible, find out if they are following strict pharmaceutical methods to produce it. Good quality Modafinil, for 50 tablets, will normally work out to be $1.50-$1.80 (USD) per 200mg. Anything that works out to be less than $1.05 per 200mg could be fake or of poor quality. This doesn’t mean to say that the more expensive it is—that it’s automatically guaranteed to be real. Not all higher priced Modafinil will equal higher quality Modafinil. So, while there are a number of things to look out for when buying, always seek to be well informed about what exactly you are buying and, from whom you are buying from.

Bad Brands

A good way to identify a trustworthy brand is whether they are certified and qualified to practice pharmacy. In an industry that is under constant and rapid change, brands which show up-to-date industry-recognized certifications are likely good brands to deal with. It’s also worth taking the time to find out things like:
  • What quality control processes do they have?
  • Do they have a CoA (Certificate of Authenticity)?
  • What is their customer service like?
  • Do they offer refunds?
  • How long have they been in business?
  • Where do they operate from
  • Are they a well recognized and recommended provider by other Modafinil users?

Recommended Dosages

When starting out with Modafinil, the recommended dosage is to be at least 50 mg per day. As Modafinil tablets are generally 200 mg each, simply start out with a tablet quarter for the first few days. This is in order to achieve best results and to avoid any shock to your system. Provided there are no negative side effects, the dosage can then be gradually increased over a period of time. Take it at least an hour before the time you actually need it. For instance, before a study period or engaging in cognitively demanding tasks.

This is so you can take full advantage of the effects. The safest maximum daily dose is generally 400 mg (2 tablets) per day. If you find yourself needing more than 400 mg per day/24 hours you may be developing a tolerance. At this point, consider taking a break from using Modafinil for at least 2 weeks before taking it again. Then, when resuming, start with a lower dosage such as 100 mg per day and gradually work your way up from there.

Dosage Guide

It is worth noting that larger intakes, such as 400 mg and above per day, don't necessarily lead to significantly greater effects. Instead, increased dosages tend to lead to the point of diminishing returns. Meaning only so much can be taken to achieve maximum effect. Clinical trials of higher level dosages, for example, have indicated that the results provided at the 400 mg dosage are not markedly improved as compared to a 200 mg dose. The studies also suggest that Modafinil is safer for prolonged periods if taken at moderate daily doses of 100-200 mg. While the effects of taking 400 mg and above per day may last a few hours longer, long-term, it could be harmful to your health. Feel free to use the table below as a guide only.

Student users of Modafinil may be one of the fastest growing markets of the smart drug today. Numerous reports and anecdotes highlight a number of reasons why students take Modafinil. As a result, increasing their ability to train for an extended period of time while also effectively reducing fatigue.

The debate regarding Modafinil use during competition has, however, been a hotly contested topic over recent years. Given its ability to greatly improve an athletes performance, it has raised concerns about its use in a competitive context. Effective as of the 1st of January 2018. According to the 2018 World Anti-Doping Agency international standard Prohibited List, the use of Modafinil is prohibited “in-competition.”
